Microenvironment and Immunology MDSC and TGFb Are Required for Facilitation of Tumor Growth in the Lungs of Mice Exposed to Carbon Nanotubes

چکیده
During the last decades, changes have been observed in the frequency of different histologic subtypes of lung cancer, one of the most common causes of morbidity and mortality, with a declining proportion of squamous cell carcinomas and an increasing proportion of adenocarcinomas, particularly in developed countries. This suggests the emergence of new etiologic factors and mechanisms, including those defining the lung microenvironment, promoting tumor growth. Assuming that the lung is the main portal of entry for broadly used nanomaterials and their established proinflammatory propensities, we hypothesized that nanomaterials may contribute to changes facilitating tumor growth. Here, we report that an acute exposure to single-walled carbon nanotubes (SWCNT) induces recruitment and accumulation of lung-associated myeloidderived suppressor cells (MDSC) and MDSC-derived production of TGFb, resulting in upregulated tumor burden in the lung. The production of TGFb by MDSC requires their interaction with both SWCNT and tumor cells. We conclude that pulmonary exposure to SWCNT favors the formation of a niche that supports ingrowth of lung carcinoma in vivo via activation of TGFb production by SWCNT-attracted and -presensitized MDSC. متن کاملEvaluation of naloxone and alum adjuvants effect in HPV vaccine on immunoediting of mice in tumor microenvironment
Background: Papilloma viruses are pathogenic double-strand DNA viruses that genotypes 16 and 18 are the cause of more than 50 percent of cancers as cervical cancer. Although vaccination is one of the best options for the papilloma cancer prevention but that is the most of world healthy problem, it is attempted to evaluate both naloxone (NLX) and alum mixture used as adjuvants together with HPV1...
متن کاملArteether Exerts Antitumor Activity and Reduces CD4+CD25+FOXP3+ T-reg Cells in Vivo
Background: Chemo-immunotherapy is one of the new achievements for treatment of cancer, by which the success of anti-cancer therapy can be increased. In vitro studies have been shown that Arteether (ARE) induces apoptosis in tumor cells, but not in normal cells. Objective: To investigate the cytotoxic and immunomodulatory properties of Arteether in-vivo and in-vitro. Methods: In this study, we ...
متن کاملListeria Monocytogenes Activated Dendritic Cell Based Vaccine for Prevention of Experimental Tumor in Mice
Background: The use of dendritic cells (DCs) as a cellular adjuvant provides a promis-ing approach in immunotherapy of cancer. It has been demonstrated that Listeria mono-cytogenes activated DCs pulsed ex vivo with tumor antigens trigger a systemic Th1-biased specific immune response and a single dose of this vaccine will cause a consider-able anti tumor immunity. Objective: The present study w...
